Every room has Unowns in it. In the long rooms, there are only one kind of each Unown that spells out the word "friend". You will find the rest of the letter Unowns in the small rooms scattered about the place.
For the "?" and "!" Unowns, you have to find every kind of letter Unown then go to Veilstone, walk down to Route 214 and head into an extremely long cave in the mountain there. You will find them in the back room of that cave right after the man who dug that cave stands.
